چکیده

Disruption of the central nervous system (CNS) barriers is one of the major pathophysiological consequences of bacterial meningitis. The increase in the permeability of the CNS barriers caused by the disruption is thought to contribute to the development of adverse neurological outcomes. We have established a method by which the CNS barrier permeability can be demonstrated by the gadolinium-enhancement ratio (GER) calculated from the T₁ weighted image (T₁WI) which is based on gadolinium-enhanced magnetic resonance imaging (GdEMRI). The present study examined the disruption of CNS barriers such as blood-brain barrier (BBB), blood-cerebrospinal fluid barrier (BCSFB) and blood-arachnoid barrier (BAB) in rats with meningitis induced by lipopolysaccharide (LPS)- or interleukin (IL)-1β. Four hours after intracisternal injection of LPS or IL-1β, severe disruption of the BAB, but not the BBB or BCSFB, was observed. This suggests that the BAB, rather than the BBB or BCSFB, plays a key role in the influx of blood-borne cells and substances during meningitis. The BAB is therefore more vulnerable to disruption than the BBB or BCSFB during meningitis in rats. In addition, GdEMRI with GER imaging analysis appears to be useful in spatio-temporal studies on the function of the CNS barriers under various physiological and pathological conditions.

Penetration of the blood-brain barrier: enhancement of drug delivery and imaging by bacterial glycopeptides

The blood-brain barrier restricts the passage of many pharmacological agents into the brain parenchyma. Bacterial glycopeptides induce enhanced blood-brain barrier permeability when they are present in the subarachnoid space during meningitis. By presenting such glycopeptides intravenously, blood-brain barrier permeability in rabbits was enhanced in a reversible time- and dose-dependent manner ...

Cyclooxygenase inhibition limits blood-brain barrier disruption following intracerebral injection of tumor necrosis factor-alpha in the rat.

Increased permeability of the blood-brain barrier (BBB) is important in neurological disorders. Neuroinflammation is associated with increased BBB breakdown and brain injury. Tumor necrosis factor (TNF)-alpha is involved in BBB injury and edema formation through a mechanism involving matrix metalloproteinase (MMP) up-regulation. There is emerging evidence indicating that cyclooxygenase (COX) in...

Biomarkers Indicative of Blood-Brain Barrier Disruption in Multiple Sclerosis

Blood-brain barrier (BBB) disruption is one of the hallmarks of multiple sclerosis (MS). It is incompletely understood whether BBB disruption is the initial MS event leading to MS lesion formation or whether it is merely a consequence of cellular infiltration in the central nervous system (CNS). The presence of gadolinium enhancing (Gd+) lesions on serial brain MRI scans is frequently used to e...

Inflammatory response after ischemic stroke: a USPIO-enhanced MRI study in patients.

BACKGROUND AND PURPOSE The intensity of the inflammatory response may be related to the volume of acute infarction. Ultra-small superparamagnetic particles of iron oxide (USPIO) may enable assessment of neuroinflammation. We aimed to assess whether the intensity of the inflammatory response might be related to the subacute ischemic lesion volume. METHODS We enrolled patients who presented wit...

Ultrastructural localization of albumin transport across the cerebral microvasculature during experimental meningitis in the rat

Injury to the blood brain barrier (BBB) is a fundamental sequela of bacterial meningitis, yet the precise mechanism facilitating exudation of albumin across the endothelium of the cerebral microvasculature remains conjectural. After intracisternal inoculation of Escherichia coli (0111:B4) lipopolysaccharide in rats to elicit a reversible meningitis and BBB injury, we utilized in situ tracer per...
